Lawtan's Rules of Comedy:

1) Timing is key.
2) Someone (or an object treated as someone) has to temporarily "suffer." (By confusion, deception, slapstick, or what have you...making suffering non-temporary leads to harassment and/or tragedy)
3) Subject material by itself isn't funny...but people's (inter/re)actions with/to subject material are funny. (hence why the "A ___, a ____, and a ___ walk into a bar" can be bigoted and cruel, or incredibly hilarious due solely to the interactions/the punchline)
4) Jokes have to be made under relate-able scenarios. If the scenario is too far-fetched/tangential/hollow/cardboard to relate to, it will likely fail.
5) Repeated jokes only last so long, and without decent setup and variation, they will die rather quickly. (Note: in turn, comedy benefits by new twists to one's expectations - new surprises)
