Forbidden Tower's condition is "no leader skills" -- you basically have six subs instead of two leaders and four subs. Fortunately, the monster health and attack power are reduced to compensate. The dungeons following it are particularly annoying because they require that all of your monsters have a certain element, but that element's orbs never appear. (This is less annoying now than it used to be -- it used to be you whole team had to have their MAIN element as that color, and now it can be the sub element.)

As for boss fights: That's a strategy thing. Bosses go rage mode at low health. This means that you should save your spike damage active skills until closer to the end of the fight. Gravity and poison should be used right away, of course, but damage multipliers (King slimes, Lu Bu, Kanna, Muse/Delgado/etc.), defense breakers (barons, Machine/Dark Golem, Top Droidragon, Shiva), and massive direct-damage attacks (Lucifer, Raging Kagato) should be saved for when you know you can spike the boss all the way down to 0% in one turn. (Of course, if you have so much spike damage lined up that you can OHKO them, then that means you use them on turn one.) This also means that when you're getting the boss close to its rage mode (usually 20% but some bosses start getting nasty as high as 50%) it can be wise to save up attacking orbs for a turn or two so you can get a stronger combo -- for example, if a single 3-match would pull the boss into rage mode, stall until you can use multiple attacking 3-matches (or 4-matches for a TPA team) to try to blast past it.

Delay effects (Echidna, Orochi) get used depending on if/when the boss uses a status ailment shield. Late-game, most bosses throw up a status ailment shield as a preemptive, so delay effects are useless, but some don't do it until their first turn so using a delay right away is ideal there. For bosses that don't use a status ailment shield at all, or for those that have short durations on their shields, saving the delay until you need it to prevent them from using super-powerful attacks is best.
